How long does it take to develop and approve a vaccine to market, regardless of safety or effectiveness? Sure many here know MUCH more about this than me. Please share your knowledge.
With the announcement in the UK that Boris Johnson plans to begin mass COVID vaccination by the winter - that’s less than 9 months of vaccine development and testing - I thought I’d look into the time it has taken in the past to achieve such a feat. Sure similar “miracles‘ are happening in your countries too.
Since it has been sometimes tricky to find exact dates for the beginning of vaccine research, I have also been using date of discovery of virus. The information below is the result of an hour or so’s research, and there is clearly much more that needs to be added.
Even after the lengths of time noted below, many of these vaccines were neither safe nor effective.
It need not be pointed out that vaccine development for COVID is a current priority to the pharmaceutical community and time is apparently of the essence, but we cannot ignore the necessity of a period of years before market, in order to properly understand the long term ‘side’ effects. Even when true, if a vaccine is deemed safe after six months testing, the company can only give assurance of safety for that six month period and no longer.
And, yes, I know that no vaccine is ever 100% safe or effective, but this will give us a rough idea of previous vaccine timelines.
EDIT: from clemaneuverers:
That can sometimes take even longer.
Polio first properly analysed in 1890... In 1951 Salk began work on the vaccine and by 1954, the vaccine was tested for its ability to prevent polio; the field trials involving the Salk vaccine would grow to be the largest medical experiment in history. Immediately following licensing, vaccination campaigns were launched, by 1957. [We know the problems associated with polio vaccines and the evidence for environmental toxins and against germ theory here...]
Since the 1970s, there has been a considerable effort to develop similar vaccination strategies for [malaria in] humans. The first vaccine, called RTS,S,was approved by European regulators in 2015.
Malaria has been around for centuries, diagnosed by 1717, and the BMGates-backed GSK vaccine was finally released in 2019. [Results? Not great - even after 7+ years of research and testing!]
Del Bigtree video:
“In November 2012, findings from a Phase III trial of RTS,S reported that it provided modest protection against both clinical and severe malaria in young infants.“,S
So, specific research must have been underway WELL before 2012.
Ebola was first discovered in 1976. Vaccine approved in 2019.
[Ebola] Vaccine development began in the late 1970s: results from a test of inactivated Ebolavaccine in guinea pigs were published in Lancet in 1980
Lyme disease was diagnosed as a separate condition for the first time in 1975...A vaccine for Lyme disease was marketed in the United States between 1998 and 2002, but was withdrawn from the market due to poor sales.
Hepatitis A was first was discovered by Steven M.Feinstone back in 1973. In the US, the vaccine was first used in 1996 for children in high-risk areas, and in 1999 it was spread to areas with elevating levels of infection.
Dengue fever has been around for centuries, ‘discovered‘ in the 1970s, diagnosed in the late 1990’s, with vaccine produced 2015.
“The bad news is it’s at the centre of a vaccine disaster story. In 2016 its world-first mass roll-out in the Philippines caused political scandal, the exhumation of children’s bodies, and an ongoing court case.
Indirectly it’s been blamed for a distrust of vaccines, muddying the waters of scientific consensus on vaccine safety, and a measles outbreak which has even reached New Zealand shores.”
Rotavirus was discovered in 1973 by Ruth Bishop and her colleagues by electron micrograph images and accounts for approximately one third of hospitalisations for severe diarrhoea in infants and children...The vaccine first became available in the United States in 2006.
In 1999, a group of scientists, including Cancer Research UK scientist Professor Julian Peto, diligently re-tested the samples and found that virtually all cervical cancer samples (99.7 per cent) contained HPV.”
In December 2014, the FDA approved a nine-valent Gardasil-based vaccine.
“Vaccine development is a long, complex process, often lasting 10-15 years and involving a combination of public and private involvement.”
Paul Offit, MD, describes the general process of making a vaccine. The College of Physicians of Philadelphia
"The AIDS vaccine has been worked on for 20 years or more, for example, and it's still not out," says Dr. Zervos. "But other vaccines have been developed more quickly than that, and our hope is that this one will be ready in 18 months, although there's always the possibility it doesn't work and you have to start over."
“In 1963, a star researcher at the pharmaceutical company Merck took a swab of his own daughter’s throat to begin cultivating a weakened form of the mumps virus. And just four years later, in record time, Merck licensed Mumpsvax as the world’s first effective vaccine against this common and contagious childhood illness.”

2020.04.22 13:24 Smooth_Imagination Asthma Under-represented? If so, Protection by Disease or by Inhaler?
Doctors are saying that asthmatics are under-represented, which is pretty surprising, you would expect an increased risk that would be fairly noticeable.
It seems very intriguing and potentially illuminating for COVID19 treatment, so the following bit of speculation I could not resist whilst we wait for confirmation of under-representation.
Is it the disease that is protective or maybe the treatment?
First, the treatments in asthma involve inhaled corticosteroids. Its been widely said that corticosteroids = death in the treatment of severe respiratory infections because they are immuno-suppresants.
But is this actually valid or simple received wisdom that slipped through?
In summary, the blanket inclusion of inhaled corticosteroids in the recent FDA label warning of an association between severe varicella infection and corticosteroid therapy without reference to dosage, and proof of immunosuppression and subsequent increased risk, dose not appear warranted. To date, no link has been established between inhaled steroids and pulmonary or systemic infections.
A more recent meta analysis says yes, but bizarely with low dose and not high dose of inhaled corticosteroids (but they were only looking at upper respiratory tract infections, and not severity of outcomes)
Moreover, fluticasone was observed with an increased risk of URTI (Peto OR, 1.18; 95% CI 1.02-1.38; p = 0.03; heterogeneity: I2 = 21%) but not budesonide, low-dose fluticasone treatment was associated with a significantly higher risk of URTI but not high dose.
So perhaps this lends support to use of inhalers as delivery for corticosteroids, because I can well imagine an asthmatic going down with symptoms of COVID19 and reaching for a puff on the inhaler, but as I understand it doctors will not give this to non-asthmatics.
We now have more more compelling information to link neutrophils to bad outcomes and so this may be of note -
Inhaled corticosteroids reduce neutrophilic bronchial inflammation in patients with chronic obstructive pulmonary disease
Inhaled corticosteroids increase blood neutrophil count by decreasing the expression of neutrophil adhesion molecules Mac-1 and L-selectin.
This seems to stop neutrophils migrating to the lungs.
What if the disease is protective. Could this be possible?
Asthma involves an increase of a protein called Pendrin, which transports thiocyanate into the respiratory lining. It's possible, although a fringe hypothesis, that thiocyanate is protective up to certain levels. An excess of thiocyanate for a long time is also bad however, but it can reduce injury caused by neutrophils when they are activated, and when otherwise there is insufficient thiocyanate available,

GameSoulz - GameSoulz Unscored ~ Unscored Some of the initial vision, gameplay and appeal may be lost, exchanged for a more streamlined, smoother experience but that doesn´t mean its worse. No, Tango Gameworks delivered a game expanding on Mikami´s initial vision, mixing it with more western ideas into a damn good game, only held back by a too foreseeable story and some minor flaws. XB1
Eurogamer - Edwin Evans-Thirlwell Unscored ~ Unscored Resident Evil 4 meets the Truman Show in an entertaining but unremarkable follow-up, held back by tepid stealth and warmed-over scares. PS4
Rock, Paper, Shotgun - Olivia White Unscored ~ Unscored The Evil Within 2 feels like something of a departure from the first game, but also an extremely fitting follow-up. Its structure, enemy design, immaculate audio production and constant tension make it one of my favourite survival horror experiences to date, and while it doesn’t push the envelope in terms of providing anything new, it focuses on what it is and attempts to provide a definitive, well-produced classic survival horror experience. It succeeds at this on basically all counts, with the minor combat quibbles doing little to dull the overall experience. PC
Ars Technica - Steven Strom Unscored ~ Unscored The Evil Within 2 is a mechanical step up from the first game in nearly every way, even if the narrative is just as disposable as ever. Buy it if that balance doesn't bother you. PC
Kotaku - Stacie Ponder Unscored ~ Unscored This game is everything I love about survival horror. There are foreboding environs to explore. Horrific monsters both large and small to slay or avoid. The sound design sends chills down the spine, from the evocative soundtrack to the anguished moans of The Lost. There is plenty of challenge, and players must choose their battles wisely and carefully manage their resources. With a shocking amount of blood and a surprising amount of heart, The Evil Within 2 slashes, stomps, and sneaks its way into the pantheon of all-time greats.
USGamer - Mike Williams Review-In-Progress ~ Review-In-Progress From what I've played so far though, The Evil Within 2 is everything I wanted from the first game. Every major complaint I had about the first game is at least addressed in some manner, though the tweaks aren't always perfect. PC
AngryCentaurGaming - Jeremy Penter Wait For Sale ~ Wait For Sale This is actually a 'Wait for Sale' and to be honest, it is very close to a 'Wait for Deep, Deep Sale'. Here's why it's not though: I actually enjoyed a great deal of what was going on with The Evil Enemy Within 2, it just felt like sometimes it was from two or three different games all mashed together. That didn't necessarily stop that fun from happening, but it really does, as you're playing it, remind you of just how cool it possibly could have been. PS4, XB1, PC
GameSpew - Richard Seagrave 100 ~ 100 / 100 2017 has been a great year for games, but for me, The Evil Within 2 has been one of the most pleasant surprises. It’s a fine example of how a sequel should be done; taking what worked and made the original such a memorable experience while building upon it and stripping out the fat. The Evil Within 2 is good old-fashioned survival horror with a little bit of an open-world bent, and it works beautifully. It’s tense, it’s scary, it’s exciting, it’s absorbing – it’s everything I hoped it would be and more. If you want something horrifying to play this Halloween, make sure it’s The Evil Within 2. XB1
Stevivor - Steve Wright 90 ~ 9 / 10 It's a solid, emotional romp filled with challenge and scares alike. It's certainly recommended. XB1
AppTrigger - Daniel George 90 ~ 9 / 10 Whether it’s the variety of worlds to explore, monsters to slay, personalities to pontificate about or the added depths of survival gameplay, The Evil Within 2 will send chills down in more ways than one. Subverting the expected and expanding upon the familiar, it builds upon its own universe and gives the player more to think about once finished. Long story short; you’re in for a hell of a treat. PC
Windows Central - Paul Acevedo 90 ~ 4.5 / 5 stars The Evil Within 2 is a frightening game, though perhaps not as pants-wettingly scary as the early portions of Resident Evil 7. The game itself is improved nicely over the original, with a great open-world structure and lots of interesting missions to take on. The actual story is extremely compelling as we follow Sebastian's journey towards redemption amid a world filled with both human and inhuman monsters. If you're looking for a lengthy and frightening adventure this Halloween season, look no farther than The Evil Within 2. XB1
Pittsburgh Post-Gazette - Max Parker 90 ~ 9 / 10 Since nobody really expected "The Evil Within" to get a sequel, expectations for "The Evil Within 2" were basically nonexistent. It was up to Tango Gameworks to show us why we should want this sequel, and they have accomplished this. It succeeds where the previous game failed, while adding more to the formula that could win over those who didn't care for the first game. For those looking for something creepy this October, or for those who want to give the series a second look, "The Evil Within 2" is worth your time.
Cheat Code Central - April Marie 90 ~ 4.5 / 5 If you are considering getting The Evil Within 2, be prepared for some slightly clunky movement and somewhat repetitive walking simulator sections. Past that, it’s a wonderful sequel to a game that was originally sort of a miss, rather than a hit. You’ll become just as invested as Sebastian is when trying to find his daughter and walk away from the experience feeling like you got your money’s worth. The Evil Within 2 is well worth the purchase, but especially those who enjoyed the first game and want something more.
Game Rant - Dalton Cooper 90 ~ 4.5 / 5 The Evil Within 2 is a significant step forward from the first game in the series, providing tighter controls, a more focused narrative, and no shortage of scares. PS4
GameZone - Cade Onder 90 ~ 9 / 10 Despite some technical shortcomings, The Evil Within 2 is an incredible horror game that should not be slept on. It’s been awhile since we’ve gotten a game like this and it shows that there’s still a lot of things that can be innovated on in the genre. Tango Gameworks took all of the criticisms of the first game to heart and built an emotional game about guilt and forgiveness. The amount of depth in the game’s story is worthy of an applause and there’s a lot of fun to be had with the intense, sweat-inducing gameplay. The Evil Within 2 is a must play for horror fans thanks to its moving plot, sandbox gameplay, and nightmare-ish world and is an essential purchase for this Halloween. XB1
Easy Allies - Michael Huber 90 ~ 4.5 / 5 stars The Evil Within 2 is a ruthless nightmare that constantly challenges. The unsettling atmosphere contributes to a sense of unpredictability, and the unrelenting combat constantly takes more than it gives. Just when you start to get comfortable, it pushes back harder. Whether it’s a boss fight, a new enemy, or the ever-shifting environment, the circumstances always keep you off balance. The Evil Within 2 encapsulates the best elements of classic survival horror with a modern touch. Written PS4
Polygon - Philip Kollar 90 ~ 9 / 10 The Evil Within 2 represents one of the starkest and most astounding turnarounds from a debut title to its sequel that I’ve ever witnessed. It’s a brilliant horror game, one that understands when to ratchet up tension and when to pull back and let you collect yourself. If the first game was a failed attempt to capture the spirit of Shinji Mikami’s classic Resident Evil 4, the sequel is a successful attempt at something much better: finding a chilling, exhilarating voice of its own. PC
GameCrate - Leo Parrill 88 ~ 8.8 / 10 The Evil Within 2 improves on the sequel in every way, while retaining the core of what made the first one special. Better gameplay, better story, and more options make this an all around excellent survival horror experience. PC
Spaziogames - Domenico Musicò - Italian 85 ~ 8.5 / 10 The Evil Within 2 is a very solid and thrilling survival horror. Despite some not so good tension breaking sandbox sections, filled with unnecessary fetch quests, this sequel is better than its predecessor: better game rhythm, better story, no more frustration and a compelling skill system. PS4
IGN Spain - Jose A. Rodríguez - Spanish 85 ~ 8.5 / 10 The Evil Within 2 is again an amazing chance to push ourselves into the darkness of fears and nightmares, a very psychological experience with an interesting plot and an incredible use of terrifying creatures. PS4, PC - Paula Sprödefeld - German 85 ~ 85 / 100 I did not want to stop playing, because even if the story of The Evil Within 2 was a bit confused, I wanted to experience the end anyway. And it's worth it, believe me. PC
Areajugones - Víctor Rodríguez - Spanish 85 ~ 8.5 / 10 The Evil Within 2 is a sequel that hooks the player from the first minute. Even though the AI has some issues and some basic enemies, the gameplay quality, its amazing visual style and a great story make this game one of the best horror experiences nowadays. PC
Atomix - Claudio Quiroz 85 ~ 85 / 100 It is said that sequels are usually the best games in a series and we can say that with The Evil Within II that happened. With a new world to explore in non linear gameplay, detective Sebastian Castellanos is ready to fight in a new nightmare in order to save his beloved daughter. Survival Horror fans must try the new game. PS4
PC Aficionado - Reace Peto 85 ~ 8.5 / 10 Evil Within 2 is a fantastic game with a compelling story and engaging gameplay. I fully recommend it to any fans of the Horror genre. PC
GearNuke - Humad Ali Shah 85 ~ 8.5 / 10 The Evil Within 2 can be genuinely terrifying at times. The game masterfully blends survival horror elements with plenty of action in between moments of silence. As a sequel, it is a bigger, better and ultimately more enjoyable experience. PS4
Hobby Consolas - Daniel Quesada - Spanish 82 ~ 82 / 100 A remarkable adventure that offers a great atmosphere, story and a solid combat. Don't expect to be scared by it, though. PS4
The Games Machine - Mario Baccigalupi - Italian 82 ~ 8.2 / 10 The Evil Within 2 improves a lot over the first title, with better graphics, more action elements and a more open level design. Nevertheless, it remains faithful to the original formula, and does an excellent job in blending narrative, gameplay and aesthetics in order to create a robust and challenging horror game. PS4
GameSpace - William Murphy 82 ~ 8.2 / 10 The Evil Within 2 is a memorable and compelling game all on its own, and one that deserves to be played by horror fans and action game fans alike. The frustrations of the first game (which have been documented by others) are non-existent here and instead, this sequel outshines its predecessor in almost every aspect. Next time, just give us a bit better story and dialog, and a more interesting protagonist. PC
Twinfinite - Zhiqing Wan 80 ~ 4 / 5 As it stands, The Evil Within 2 is held back by a forgettable story and annoying AI issues that would probably put a serious damper on your gameplay experience if you were playing on Nightmare difficulty. And yet, it also does exactly what a horror game should do, and it does it well. Despite its flaws, The Evil Within 2 is a thoughtfully constructed horror rollercoaster ride that strikes a delicate balance between giving the player the power to fight back, and also making them realize that sometimes, all those guns and gear won’t save you from the terrors you can’t understand. PS4
IGN Italy - Biagio Etna - Italian 80 ~ 8 / 10 The Evil Within 2 is definitely one step ahead of its predecessor: a rich, deep and long-lived horror survival. The level of difficulty has been considerably softened. If this makes the title more accessible, on the other hand, he denies that authoritative uniqueness of the title that inspired it. PS4
Digital Trends - Steven Petite 80 ~ 4 / 5 stars The Evil Within 2 overhauls every aspect of the original to create a surprisingly adept sequel that makes us a hungry for more. Both gunplay and stealth have been refined and lend themselves well to the restrained open world format. Its surprisingly heartfelt narrative and many twist and turns round out the experience. PS4
Gamespot - Alessandro Fillari 80 ~ 8 / 10 Though there's some occasional technical hiccups that result in some particularly frustrating moments and weird pacing issues, this horror sequel elevates the tense and impactful survival horror experience in ways that feel fresh and exciting. What this cerebral horror game does isn't totally new, but it rarely feels routine, and offers plenty of surprises. Coming in at a lengthy and surprisingly packed 15-hour campaign, the sequel does an admirable job of ratcheting up the tension and scares when it needs to, while also giving you the freedom to explore and proceed how you want. It's a tough thing to balance, but The Evil Within 2 does it remarkably well, and in a way that leaves a strong and lasting impression after its touching conclusion. PS4, XB1, PC
IGN - Lucy O'Brien 80 ~ 8 / 10 The Evil Within 2 gives you more options to play your way in a much bigger survival-horror playground than the original. Although a disjointed plot and weak protagonist make it slow to get started, being pursued through an intriguingly bizarre world by plentiful and disturbing monsters kept my palms sweaty and my heart in my throat thanks to a strict adherence to the most important survival horror rule: you should always be on the back foot. XB1
Worth Playing - Cody Medellin 80 ~ 8 / 10 The Evil Within 2 is a good survival horror game for genre fans to sink their teeth into. The story makes more sense this time around, and the atmosphere delivers the right amount of unease, which is what will keep players plugging away to the end. The gameplay has been tweaked to be easier, but it's not to the point where you can go through without much trouble. Despite the presence of a few technical issues, TEW2 looks and sounds great, and it's certainly worth your time, even in this incredibly packed gaming year. PS4
Gameblog - Jonathan Bushle - French 80 ~ 8 / 10 The Evil Within improves the game mechanics by adding the exploration of a few open zones. A very good game for horror fans! PS4
CGMagazine - Joel Couture 80 ~ 8 / 10 The Evil Within 2 is horrifying delight, using a feeling of vulnerability to make players appreciate every moment they’re still alive. PS4
PlayStation LifeStyle - Paulmichael Contreras 80 ~ 8 / 10 The Evil Within 2 has arrived just in time for the spookiest holiday of the year. While the difficulty level has been ratcheted down noticeably in the name of accessibility, the harder modes should suffice for those looking for a challenge. Creepy environments are a staple of this series, and there are even a few jump scares thrown in for good measure. A much more focused story will drive you to want to play for “just one more chapter,” while a decent selection of side missions will also keep you occupied for the duration. Still, by incorporating a lot of the more traditional elements of open-world games, such as a regular map, waypoint system, and lengthy cutscenes, the game has lost some of the abstract charm seen in the original. This should not act as a deterrent, however; horror fans have found the best game to play this Halloween. PS4
Gamers Heroes - Johnny Hurricane 80 ~ 8 / 10 The Evil Within 2 took what The Evil Within did and improved upon it in every way. If you need to scratch the survival horror itch, The Evil Within 2 is an easy sell. PS4
Impulsegamer - Sean Warhurst 80 ~ 4 / 5 John Johanas has masterfully taken up the directorial reigns and given us a game that, while it shares many trace elements of DNA with its predecessor, has more than enough of a distinct identity in order to separate it from the pack. PS4
AusGamers - Kosta Andreadis 80 ~ 8 / 10 The tension and intensity can be felt throughout both the open-world sections, where you can choose where to go, as well as those where you’re simply walking down a hallway filled with framed pictures highlighting truly chilling acts of violence. It’s a thrill ride quite unlike any other, and as a survival horror release it will leave you shaking or at the very least on the verge of needing a nice long vacation. XB1
GameSkinny - Ty Arthur 80 ~ 8 / 10 stars Bringing with it some of the best parts of Residen tEvil and Silent Hill, The Evil Within 2 does a lot of things right. XB1
Telegraph - Sam White 80 ~ 4 / 5 stars The game's greatest strength is avoiding the pitfalls of the first; removing frustrating boss fights and lessening the amount of forced stealth sections.
Press Start - James Mitchell 80 ~ 8 / 10 The Evil Within 2 is an enjoyable, psychological and somewhat emotional trip that I probably won't forget. As a sequel to it's predecessor, it takes things in a rather different direction and tries several new things to mixed results. Nonetheless, The Evil Within 2 is a thrilling and tense experience that'll appeal to most fans of the original and newcomers looking for a harrowing experience they've probably not had before.
The Outerhaven Productions - Keith Mitchell 80 ~ 4 / 5 stars The Evil Within 2 does a good job of finishing the original story. Seb gets reunited with his daughter and overcomes his guilt. That said, the game revisits multiple events from the first game. This assumes you've played it and if not, you're left following scraps of content from the game. Outside of the story, the game is quite different from the first. It's a good follow-up, even if the horror takes a back seat to the action. Thankfully the game also addresses the performance issues from the first game and it runs like a champ (if you have the hardware). A solid entry for those new to the survival horror genre. While veterans may be a bit disappointed as the game puts more focus on combat than providing the scares. Still, fans of the first game will eat this up and come back for seconds. PC
Reno Gazette-Journal - Jason Hidalgo 80 ~ 8 / 10 The Evil Within 2 returns with a world more open to exploration plus plenty of extra polish to the familiar Resident Evil formula. Admittedly, the familiar gameplay might give veterans of the genre a been-there, done-that feeling. The addition of more open-world elements, however, improves on the old RE-style approach and the third-person perspective will be welcome to folks who didn’t quite like Resident Evil 7’s switch to a first-person perspective. If you love the first game or Shinji Mikami’s Resident Evil titles, this is worth taking a stab at the old bathtub once more, pun so totally intended. XB1
PC Gamer - Joe Donnelly 80 ~ 80 / 100 An intense and thrilling psychological survival horror sequel that improves on its forerunner in almost every way. PC
PlayStation Universe - Garri Bagdasarov 80 ~ 8 / 10 The Evil Within 2 is a great addition to the survival horror genre. It improves in a lot of aspects from its predecessor, but also makes some sacrifices that made the original so memorable from a horror standpoint. The Evil Within 2 is a damn good game and any fan of the genre should give it a shot. PS4
GamingBolt - Shubhankar Parijat 80 ~ 8 / 10 The Evil Within 2 isn't without flaws, but it manages to deliver a great blend of horror and action. PS4
Game Informer - Suriel Vazquez 78 ~ 7.8 / 10 The Evil Within 2 is a solid horror game that expands the scope of the series to extend out of the shadow of a titan in the horror genre, even if it sometimes leans heavily on borrowed ideas. Its progression and difficulty curve create a satisfying loop that repays resourcefulness and strategy, but its set pieces and structure don’t build enough on the many ideas they borrow to make them feel new or interesting. The result is a ride that offers some fun thrills in the moment, but I don’t think I’ll lose too much sleep over these particular nightmares. PS4
GamesBeat - Stephanie Chan 75 ~ 75 / 100 If you’re a horror buff, you’ll likely be disappointed because there aren’t that many unique monsters in the game. It’s not even scary, to be honest; most of the tension and fear comes from the lack of ammo. All in all, it’s not a bad game, but it’s not a great game either. You’ll probably have fun shooting the zombies and checking out the occasionally cool surreal locales, but that’s it.
COGconnected - Paul Sullivan 75 ~ 75 / 100 Despite all that up and down, I had (and still am having) a really freakin’ good time playing The Evil Within 2. While it didn’t scare me as much as I wanted it to, it packs plenty of tense moments and interesting situations into its somewhat inflated length and is definitely worth investigating for its atmosphere alone. A ‘more than the sum of its parts’ situation, The Evil Within 2 should be firmly on the radar of anyone who really loves Resident Evil 4, and even players just looking for some spookiness this Halloween. PS4
GamesRadar+ - Lucas Sullivan 70 ~ 3.5 / 5 stars Though it doesn't outdo its predecessor, The Evil Within 2 delivers another fun, challenging, tense horror headtrip that should delight fans of the first game. PS4
Game Revolution - Paul Tamburro 70 ~ 3.5 / 5 stars There's plenty tucked away in The Evil Within 2 that will both appeal to both fans of the original and new players, with it presenting a mix of Mikami's best ideas and John Johanas' new direction. It's certainly not what I was expecting, but in a good way. PC
Hardcore Gamer - Jordan Helm 70 ~ 3.5 / 5 The Evil Within 2 is a more coherent and purposeful journey, strengthened by its voyage into non-linear level design, while at the same time reinforcing the foundation with which the original’s gameplay often shined through on. That’s not to say the sequel doesn’t throw up a few too many unfair fail states — be it in and out of stealth mode — not to mention the occasional performance mishaps that show up. It’s during combat where the biggest flaw unfortunately shows itself, ultimately preventing the sequel from achieving a higher level of success. Where it may falter on making combat feel fair, it’s the visual side of things that impress and surprisingly so. It’s this degree of careful approach to design that does just enough to salvage The Evil Within 2 as a fulfilling step-up deserving of a second visit. PS4
Xbox Achievements - Richard Walker 70 ~ 70 / 100 The Evil Within 2 will certainly hold your attention during its 17 chapters, with flashes of brilliance sandwiched in among the game's various flaws and less memorable sequences. Ultimately, as a sequel, it doesn't quite deliver, but nonetheless, The Evil Within 2 is a survival horror that's just about worth persevering with. XB1
Push Square - Joey Thurmond 70 ~ 7 / 10 The Evil Within 2 has fine-tuned its solid mechanical base and visuals, and even crafted some big improvements to areas such as exploration and character direction. Yet it lacks creativity and diversity with its level design and bosses. The game may be more presentable and refined than its predecessor, but it's also lost some substance in the process. PS4
We Got This Covered - Toddy Rigney 70 ~ 3.5 / 5 stars When compared to other games in the horror genre, The Evil Within 2 seems a few steps behind its contemporaries, both in terms of storytelling and execution. However, what it lacks in genuine scares and creepiness it makes up for in atmosphere and gore, especially during the final chapters. PS4
Destructoid - Raymond Porreca 70 ~ 7 / 10 The Evil Within 2 often feels uneven, but that's probably the point. Even if it isn't, there's enough blood, guts, and horror to keep your heart racing. PS4
Attack of the Fanboy - Lewis White 60 ~ 3 / 5 stars The Evil Within 2 is just like its predecessor: a mixed bag. While it had the potential to be something absolutely brilliant, Tango Gameworks has attempted to both cram too much in and offer too little at the same time. It’s at its best when its following the same structure as the original; its linear campaign offers far more content and polish than its barebones open world and has obviously had a lot more effort poured into it. That said, if you still want a Resi-style survival horror game with some of the best art direction in the genre, The Evil Within 2 has you covered. XB1
Slant Magazine - Ryan Aston 60 ~ 3 / 5 stars Because The Evil Within 2 is otherwise successful at being a potent action-horror experience, such design failures are bound to stick with the player all the way through to the final act XB1
Shacknews - Chris Jarrard 60 ~ 6 / 10 Castellanos pushes through the twisted world in the hunt for Lily, encountering new faces, old faces, disgusting nightmare faces, and a semen monster. By the end of it all, I couldn't remember half of what happened and had no desire to explore STEM for a second time on a higher difficulty. Reducing the amount of bullets is not going to make the experience more compelling. PC
VideoGamer - Alice Bell 50 ~ 5 / 10 Some monstrously lovely set pieces are let down by a bit too much padding, some broken mechanics, and an overwrought story. PS4
Metro GameCentral - GameCentral 40 ~ 4 / 10 A worse game than the already flawed original, whose poorly crafted action is as ineffective as its clichéd attempts at horror. PS4
TrustedReviews - Phil Iwaniuk 40 ~ 2 / 5 stars If it's possible to preserve a sense of tension and genuine horror in an open-world setting, The Evil Within 2 hasn't discovered how. In an attempt to introduce a more contemporary gameplay style than that of Shinji Mikami's classics, this game ends up diluting its best qualities by filling your time with tedious milling about and resource-gathering. PS4

2017.03.07 18:04 izveniklickov Bitcoin Drops on PBOC, Waits ETF

Another exciting week in the crypto arena. Both bitcoin and litecoin clocked some gains since our last weekly update. But will these gains stick?
Bitcoin Drops on PBOC Comments
Bitcoin prices dropped from $1,255 to a low of $1,170 today on comments by PBOC’s Zhou Xuedong. The official said that while bitcoin exchanges will not be forbidden, strict regulations will be applied to the sector. The PBOC will undertake an observation period and may even close non-complying exchanges. Zhou Xuedong is the director of the PBoC’s Business Administration unit and was the person in charge of the audit conducted on bitcoin exchanges earlier this year.
This tone is the strongest yet to date from the People’s Bank of China so it’s not surprising to see the price fall a decent amount. However BTC/USD has recovered about half of the losses already and is quoted at $1,226 right now. Chinese prices remain suppressed as bitcoin withdrawals are still not possible. One coin is selling from $1,150 t0 $1,165 on the Big Three, a discount of over $70 dollars from USD exchanges. Imgur Read more:

For this specific implementation the goal is to create decentralized disruptive products and services alternatives, targeted at high value industries while providing the highest value possible to end users of such services.
Evolution : platform & name change
SuperDAO organization came about as a governance need at the same time frame as the Pokereum dapp initiative back in late 2013. As such, both projects have undergone considerable platform and branding changes. Moreover, SuperDAO’s core team has had ample time to iteratively innovate over DAO issues such as; tyranny of the majority, voting quorum or voter apathy issues and DAO product development efficiency risks. Some of the viable solutions invented by superDAO to solve the described problems are; dynamic voting, Internal core reputation voting, structured core contributions and bounty-impact based, mission aligned, arbitrary contributions.
Branding and Platfrom changes :
Platform :: NXT - > DAO (Eth) / Dapps (Eth /Etc / Rootstock) Dapp Branding :: Nxtpoker -> Pokereum Oganization Evolution :: NxtPoker DAO -> Pokereum DAO --> SuperDAO
Governance mechanism:
The SuperDAO multi-tiered organization onion model provides for a cryptographic tokens layer and an internal reputation-based layer called "Merits". The cryptographic tokens sovereignly governs all affairs, while internal Merits provide for efficient management and development. At the center of the layers is the consititutionalDNA which carries the mission, vision and contractual values of the Decentralized Autonomous Organization providing a system of checks and balances wrapped up in a simple modular hub contract called consensusX.
Bootstrapping the Superdao is a delicate process necessary to gradually bring the DAO from conception to full crowd governance. SuperDAO utilizes three defined smart contract enforced states to indicate its stage of development evolution and governance modes:
Pokereum Decentralized Provably fair Poker is the first Superdao Dapp. It Provides a polished user experience alongside a provably fair solution to the mental poker problem using smart contracts as a component to a complex, multi-faceted solution. The research and draft papers were first made available before the advent of the Ethereum platform. While some teams are attempting to solve similar issues using cryptographic primitives and outdated techniques, Pokereum is uniquely positioned as leader and pioneer and provides a solution that leverages cryptographic properties of cards games, smart contracts in conjunction with client side technology. Its updated solution "Shadow Poker" will provide any other similar project a viable solution for integration. the UX, state & event communication layer uses real time peer to peer telehash framework and is complete with poc 1( Shadow Poker is scheduled for release and review shortly after superDAO's membership driven crowdfunding is completed.

The second SuperDAO Dapp; Dappsery is an advertising marketplace Dapp which uses Ethereum payment channels to enable timely micro-payments ands gives publishers and advertisers ability to interact directly without additional third party costs. It also provides for a fraud deterrent solution which aspires to reduce click fraud to a point beyond concern. Dappsery has social and contextual advertising verticals in its cross hairs, but is currently focused on display advertising with a prototype in the early stages of development. In general, the marketplace will be positioned to disrupt a massive $500 billion industry.

Gains and losses of government forces and militias.
The government forces have conducted another offensive today and managed to capture a fair amount of villages, those are the following: Al-Sabiyaha, Bagtos, Gnamiya, height 476 and Han Al-Zhu. The settlements of Ara, Al-Hawr, Duveyrika and Tiger hill remain contested, with clashes going on between JaN and the government forces. The capture of Khan Al-Zhu means the government forces managed to capture the exchanger that connects the western part of Latakia with its eastern part, this means the highways can no longer be used by the rebels. The advancements of today are major as they impair the movement ability of the rebels.

map Map made by PetoLucem and posted here all credit goes to him.
Al-Qaratayn is starting to get surrounder by government forces by atleast two sides, there were reports of government troops approaching from the south and from the west. Al Qaratayn is a key city in this conflict as its capture will enable the government forces to recapture Palmyra. Palmyra is on the road that leads to Deir-Ezzor.
The government forces are standing back for now on the other fronts, no specific captures have been reported. It is known that they are preparing an offensive in eastern and southern Aleppo but the starting date for this offensive is not yet known.
Relatively quiet day so question for today: What do you think Turkey's plan is in Syria?
Top 5 posts of the day:
  1. Suleiman Assad sentenced to 20 years in Colonel Hassan Sheikh murder
  2. Turkey bombs Kurdish headquarters northern Syria - ARA News
  3. US (Kerry) says no fundamental delay to Jan. 25th talks. Russia (Lavrov) says no intention to delay. Turkey (Davutoglu) says Russia insists on "terrorist group" YPG in opposition. Iran (Zarif) says 10 Riyadh opposition members are Al Qaeda. UN (de Mistura) says "I will tell you on the 24th."
  4. Russia deploys troops in Qamishli to examine the airfield and other facilities
  5. French special forces operating against ISIS on the Mosul Front in northern Iraq -
If anything in this listing is off or incorrect, do tell me and I will adjust.
I could have missed any captures or retreats from both the government forces and the insurgents, if this is the case, please notify me and I will add or remove parts of the summary.
If you have problems with sources or feel like some sources should have a pro-X tag, do tell me and I will modify accordingly.

Just wanted to know what, in your opinion, is the biggest barrier at the moment between researchers and the next breakthrough in cancer prevention/cure? I've seen you mention different types of cancers and not yet understanding the immune system completely, but is/are there other factor(s)? I think that the biggest barrier is investment in clinical trials. Clinical trials are our only tool that can tell us if something works or not. Unfortunately, they are also very, very, very expensive. Right now, the high costs of clinical trial pushes industries to invest only in a safe way that is in very frequent diseases and where they know with high confidence that it is gonna work. Moreover, due to safety regulations we know nothing about drugs that we already have and their efficacy on other types of disease: if a drug is approved for EGFR-mutated breast cancer, you cannot use it on EGFR-mutated lung cancer, even if we are talking about the same target (EGFR) present in two different cancers. A health insurance company won't cover the costs of something that is not approved. Of course, there is a high risk to lose your money. Similarly, industries are not always happy about this off-label treatments because there is high chance that it won't work (bad publicity for the drug). I think that reshaping the way we finance clinical trials and making it easier to try off-label treatment would help a lot in this regard... Maybe better treatment schedules are under our nose and we can't notice it, because there is too much fear to try and lose money.
Follow up question to this. How soon/important do you see stem cells and organ creation (such as livers, etc. where they create the frame and line it with stem cells or grown from cells) in bringing down the immense cost to trials? There would be a lot less risk and I would imagine cost to lets say grow a tumorous liver and use drugs on a grown organ but would data from a trial such as this be as viable without the rest of the human body (such as the lymph system)? Growing isolated organs would be certainly an advance. However, it would not address the problem of drug toxicity in other districts of the body. We cannot afford to give up on trials even with such advancements. What these technologies would do is to filter the best candidates to promote to clinical trials. So hopefully we'll have better pre-clinical research, more predictive about the success of a drug, when tested in clinics.
How fast is actually a ki67 of 90%? A Ki67 of 90% is very high, but it doesn't tell about the speed of growth. Actually, it means that 90% of cell are probably going to divide but it doesn't tell us when. Anyway, we can assume it indicates a fast growth rate and that's very good sign that it didn't spread.
Dont the body make its own glucose anyway? I suggest in other comments to watch this video from Stanford University: Link to It covers the topics you mentioned in an honest way. You are right in thinking that many of the "popular" food recommendations are not really scientifically founded. Generally speaking, I think that a healthy and varied diet (as you mentioned) is the best option and this would include also things like sugar and meat (if one wants), but, obviously, in the right amounts. There are some indications for cancer patients that keeping a balanced diet can actually improve therapy outcome, but again, this has nothing to do with eating particular foods or totally avoiding others. I encourage you to talk to a nutritionist, which can advice you about your diet as a cancer patient.
Regarding metformin: it has been noticed that people on anti-diabetic drugs have lower risk of developing some cancers. It is not known whether this depends on their condition or this is valid in general. Anyway, this drug would not work by simply cutting sugar intake, it has a more complicated, unrelated mechanism.
Thanks for a good answer! See I did not know that about ki67, because all I have heard is that it say something about how fast the cells divide. Looking at it that way makes a lot of a diffrence (in the mind of a patient). Took course with my SO, about breast cancer and the time after treatment. Tried getting some answers from the oncologist after the lecture, but he could not really answer me. Have talked to sevral nutritionists, but only one time by doctors appointment. I kinda lost alot of weight during treatment, BMI 18,someting now. Thanks for the answer about Metformin, because I did not get a hang of why it would work. I have my oncologist on the case to see if I can get it. Just want something preventive, like the other breast cancers have hormone therapy and herceptin. Do you think we know the long term affect of all the additives that we put in our food today? I had a course on food additives during my masters' degree... And actually no, we don't know the long-term effects for most of them. We kinda of know the acute effects and there are regulations about the maximum dose allowed, but basically you can't control if people by consuming different foods pile up a high dose of additives. More importantly, we know very few things about the interaction between them, which is another problem when it comes to real life and you get different additives from different foods. Btw, I want to make it clear that not all additives are bad! And that current regulations keep safety for consumers at the first place. The fact that we don't know so many things is technical and not by lack of will: it is difficult to study long-term effect and even more if an additive has been introduced only relatively recently on the market.
A few years ago my mother was diagnosed with myelodysplastic syndrome and earlier this year she died of acute myeloid leukaemia. In myelodyplasia and even more in leukemia, one of these cells starts dividing out of control. The offspring of this renegade cell tries to produce mature blood cells, however it fails at it and accumulate at an immature stage. The few mature cells that are produced are mostly not functional. Eventually, the bone marrow is engulfed with leukemic cells that divide without really producing blood cells.
Additionally, while we are in different countries I would like to express my gratitude for the studies and work you are doing to help others. Thank you. I am sorry for your loss, I worked for long time helping in the diagnosis of diseases like myelodysplasia and acute leukemia, I feel very much for situations like this.
When can I just take my BEP chemo drugs in pill form? I appreciate your effort in cancer research, thanks to you and people like you, my treatment seems to be being as effective as it should be and with just a bit more luck and treatment I'll get to walk away from this hideous monster. A pill contains a tiny amount of drug that has to survive the destructive action of your stomach and intestine, get released, being absorbed, get in the blood stream and go everywhere without harming other tissues in the meanwhile. In so many steps it is difficult to predict how much will end up in the tumor in the end or that it won't do any damage in the process. The cannula is much safer and offers more control. Chemo are very destructive drugs, if you injected all of it in one place, they would destroy the tissue before getting spread. Therefore, they are injected close to the heart, so that they can be pushed immediately all over the body, reducing the harm by "diluting" it in the blood.
Good luck with your treatment! I want to propose to stop thinking about cancer as a battle against a monster or an enemy, but rather as a journey. A journey that transforms you and your way of seeing the world. In the end you'll be much stronger, much happier, much more at peace with yourself. What do you think about this idea?
Also I hope you wont mind i ask a quite stupid question...isnt the best way to cure cancer is to remove the tumor from the body? so for example if someone have lung cancer and not spread to other parts of body, simply remove the lung from the patient and implant another lung will be the best way to deal with cancer? Or if someone have cancer in their leg or arm just simply remove it and the patient is 'cancer free'? It isn't stupid. When possible cancer is removed from the body thanks to surgery. But there are two main problems: first of all, surgery cannot reach all parts of the body. Removing an entire organ would in principle do (leaving aside all the ethical issues), but an organ transplant is way too much and it is not easy at all to find a compatible donor in time. Anyway this brings us to the second reason: cancer can spread to other organs. Even when it is very small, single cancer cells can invade and form tiny colonies in other organs, which will become metastasis. We cannot detect them if they're too small. So, removing the big Primary cancer does not do anything for the metastasis. Moreover, if you remove the originary cancer, the metastasis will thrive because there will be more resources available.
Sorry if you think my question is too stupid to However, surgery IS the best way we have to cure cancer. And this is why if cancer is caught in time (read here: before it has spread) surgery can efficiently remove the cancer and the patient is cured.
What is the chance of metastasis to happen? Or is just simply a luck issue? The method used or the doctor ability does not affect it? Some tumor types metastasize more often than others and this depends on their cancer genetic constitution and time. Even if in the laboratory we can very often find ways to predict occurrence of metastasis in patients, in real clinical settings we are still not able to do it.
The method used as a therapy or doctor ability have no influence at all on chance of developing a metastasis.
Why do elephants have such low cancer rates, and what can we learn from them that might apply to humans? Animals in the wild have apparently low cancer rates only because they are less likely to survive to old age and our statistics is mostly based on occasional reports. However, it is true that elephant and big animals are composed by a huge amount of cells and we would expect an increasing chance to develop cancer with animal size. This is known as Peto's paradox. There is a very new study about it which basically tells mechanisms are in place to suppress cancer growth in these big animals. Elephants, for example, have multiple copies of a gene called p53. P53 is nicknamed "the guardian of the genome", it is probably the signle most important gene put in place between us and cancer. Having more copies of it decreases the chances of disrupting its function completely.

2015.01.07 00:46 ChrisPlays Group Coaching for NA and EU!

Hello everyone, I have seen the recent post about the idea of making a 5v5 of people wanting to get better at Counter Strike.
I have taken this into thought and found that there will be quite a large interest into events like this.
Anyway to make this happen we are looking for 10 silver to gn players who are willing to learn, 2 casters to cast the game that also have experience operating the stream and controlling the camera in game.
Then I am looking for 4 more coaches preferably ranked LEM+ to help give a analysis after the game. IF you are any of these people who want the roles of Caster or Coach add me @
If you are a silver wanting to get better I will randomly choose 10 from the comments SO MAKE SURE YOUR STEAM LINK IS POSTED!
Hope this will help the community, The Mods of /AdoptASilver
EDIT: Still trying to figure out a date for this, if anyone also has a server that is good for both NA and EU then comment on my steam profile and I will add you!
